Ring star Pacquiao, 37, will fight Timothy Bradley for a third time on April 9 in Las Vegas.
The Filipino had been considering a clash with Brit Khan.
But the Bolton fighter has missed out on the lucrative scrap.
Khan, 29, was also snubbed by Floyd Mayweather Jr in the last two years.
The former light-welterweight world champion now looks set to face British rival and IBF welterweight title holder Kell Brook next summer.
Brook's promoter Eddie Hearn has already provisionally booked Wembley Stadium on June 4 for the clash.
