The Chelsea striker was been slapped with a three-game ban for violent conduct after his clash with Arsenal defender Laurent Koscielny last month.
He has served two of those games with the third coming this weekend when Chelsea face Southampton on Saturday.
He is also suspended for Spain's game against Luxembourg next Friday but could have played against Ukraine on the Monday.
But Del Bosque decided against calling him up at all when announcing his squad today.
“What he did was unedifying, and that is why he was punished,” said Del Bosque.
“Of course, I didn’t like what he did."
He added: “He can’t play the first game through suspension and we’ve decided not to bring him.
“He hasn’t played badly and it’s not on merit.”
Costa, 26, has scored just twice this season for Chelsea, in the wins over West Brom and Maccabi Tel Aviv, and was heavily criticised for his part in Gabriel's sending off at Stamford Bridge.

Gabriel later had his ban rescinded and Costa was charged despite Mike Dean failing to send him off during the game.
His form for Spain has also been an issue. He has scored just once in nine caps, against Luxembourg in a 4-0 win, since naturalising and the yellow card he picked up in the 1-0 win over Macedonia ruled him out of their next Euro 2016 qualifier.
